Themonoclonalantibody20/70recognizesthemousereceptorforC5a(CD88).ThemouseanaphylatoxincomplementfactorC5a,asplitproductofC5,isoneofthemostpotentinflammatorychemoattractantsandinteractswithtwoC51receptors,C5aRandC5L2.MostoftheC5aeffectsaremediatedthroughC5aR.C5aRisknowntobecrucialintheinitiationofacuteinflammatoryresponses.C5areceptorisaseventransmembraneGTP-binding-protein-coupledreceptorthatbelongstotherhodopsinsupergenefamily.The45kDaC5areceptorisexpressedbyleukocytessuchasneutrophils,eosinophils,basophils,monocytes,dendriticcellsandmastcells.C5aRcanbeexpressedonbothimmuneandnonimmunecells.NonimmunecellsexpressingC5aRincludevascularendothelialcells,cardiomyocytesandbronchialepithelialcells.SignalingofC5aRinvolvesintracellularcalciummobilizationandactivationofamongothersproteinkinase-B(PKB)andproteinphospholipaseD(PLD)pathways.HighC5aRexpressionlevelshavebeenassociatedwiththepathogenesisofmanyinflammatoryconditions,autoimmuneandneurodegenerativediseaseslikerheumatoidarthritis,multiplesclerosis,atherosclerosisandinflammatoryboweldiseases.InvivoblockadeofC5aRgreatlyreducesinflammatoryinjury.Themonoclonalantibody20/70hasbeenusedinfunctionalstudiesandiscapabletopreventthebindingofC5atoitsreceptor.
